磷酸化 STAT3（Tyr705）检测原理

磷酸化 STAT3（Tyr705）检测方法用于测量 STAT3 在 Tyr705 位点磷酸化的情况。与蛋白质免疫印迹（Western Blot）不同，该检测方法完全基于微孔板，无需凝胶、电泳或转膜步骤。磷酸化 STAT3（Tyr705）检测使用两种标记抗体：一种标记有供体荧光团，另一种标记有受体荧光团。第一种抗体经筛选后能够特异性结合蛋白质上的磷酸化基序，第二种抗体则可识别该蛋白质，且不受其磷酸化状态的影响。蛋白质磷酸化会促使两种标记抗体形成免疫复合物，使供体荧光团与受体荧光团紧密靠近，从而产生荧光共振能量转移（FRET）信号。该信号强度与样品中存在的磷酸化蛋白质浓度直接成正比，并且在无需洗涤的检测形式下提供了一种评估蛋白质磷酸化状态的方法。
